Uncertain significance for Herpes simplex encephalitis, susceptibility to, 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_145725.3(TRAF3):c.962G>A (p.Arg321Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 321 of the TRAF3 protein (p.Arg321Gln). This variant is present in population databases (rs148461790, gnomAD 0.07%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with TRAF3-related conditions. ClinVar contains an entry for this variant (Variation ID: 569056). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
