Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.7312G>T (p.Asp2438Tyr), citing Ambry Variant Classification Scheme 2023: The p.D2438Y variant (also known as c.7312G>T), located in coding exon 13 of the BRCA2 gene, results from a G to T substitution at nucleotide position 7312. The aspartic acid at codon 2438 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
