NM_006231.4(POLE):c.2374A>G (p.Lys792Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K792E variant (also known as c.2374A>G), located in coding exon 21 of the POLE gene, results from an A to G substitution at nucleotide position 2374. The lysine at codon 792 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
