This variant lies in the DNAH5 gene (transcript NM_001369.3) at coding-DNA position 2250 through 674 bases into the intron immediately after coding-DNA position 2259, deleting this region. Submitter rationale: This variant has not been reported in the literature in individuals with DNAH5-related disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in DNAH5 are known to be pathogenic (PMID: 11788826, 16627867). For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (ExAC no frequency). This sequence change removes the last 10 nucleotides of exon 15, and affects a donor splice site in intron 15 of the DNAH5 gene.
