Uncertain significance for Distal hereditary motor neuropathy type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_205836.3(FBXO38):c.2905A>G (p.Ile969Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FBXO38 gene (transcript NM_205836.3) at coding-DNA position 2905, where A is replaced by G; at the protein level this means replaces isoleucine at residue 969 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 894 of the FBXO38 protein (p.Ile894Val). This variant is present in population databases (rs373620719, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with FBXO38-related conditions. This missense change has been observed in at least one individual who was not affected with FBXO38-related conditions (Invitae). ClinVar contains an entry for this variant (Variation ID: 567974). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
