Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001211.6(BUB1B):c.1919C>T (p.Pro640Leu), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BUB1B gene (transcript NM_001211.6) at coding-DNA position 1919, where C is replaced by T; at the protein level this means replaces proline at residue 640 with leucine — a missense variant. Submitter rationale: Variant summary: BUB1B c.1919C>T (p.Pro640Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-05 in 251466 control chromosomes. To our knowledge, no occurrence of c.1919C>T in individuals affected with Mosaic Variegated Aneuploidy Syndrome 1 and no experimental evidence demonstrating its impact on protein function have been reported. Two submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Both submitters classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
