Pathogenic for Hereditary diffuse gastric adenocarcinoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004360.5(CDH1):c.376_382dup (p.His128fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 376 through coding-DNA position 382, duplicating 7 bases; at the protein level this means shifts the reading frame starting at histidine residue 128, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in CDH1 are known to be pathogenic (PMID: 15235021, 20373070). This variant has not been reported in the literature in individuals with CDH1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.His128Profs*42) in the CDH1 gene. It is expected to result in an absent or disrupted protein product.
