NM_024426.6(WT1):c.1086C>G (p.His362Gln) was classified as Uncertain significance for Wilms tumor 1; Drash syndrome; 11p partial monosomy syndrome; Frasier syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with WT1-related disease. This sequence change replaces histidine with glutamine at codon 357 of the WT1 protein (p.His357Gln). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and glutamine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
