Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_023036.6(DNAI2):c.1153G>A (p.Asp385Asn), citing Ambry Variant Classification Scheme 2023: The p.D385N variant (also known as c.1153G>A), located in coding exon 8 of the DNAI2 gene, results from a G to A substitution at nucleotide position 1153. The aspartic acid at codon 385 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
