This variant lies in the WAS gene (transcript NM_000377.3) at coding-DNA position 565 through coding-DNA position 566, replacing the reference sequence with AG; at the protein level this means replaces proline at residue 189 with arginine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with arginine at codon 189 of the WAS protein (p.Pro189Arg). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and arginine. This variant has not been reported in the literature in individuals with WAS-related disease.
