This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 1081, where A is replaced by G; at the protein level this means replaces threonine at residue 361 with alanine — a missense variant. Submitter rationale: Variant summary: NBN c.1081A>G (p.Thr361Ala) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251392 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1081A>G has been reported in the literature in a cohort of individuals referred for hereditary cancer genetic testing (Tsaousis_2019). This report does not provide unequivocal conclusions about association of the variant with Nijmegen Breakage Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. Four submitters classified the variant as VUS while one classified as benign. Based on the evidence outlined above, the variant was classified as uncertain significance.
