Uncertain significance for Osteogenesis imperfecta type I — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000088.4(COL1A1):c.853C>G (p.Pro285Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the COL1A1 gene (transcript NM_000088.4) at coding-DNA position 853, where C is replaced by G; at the protein level this means replaces proline at residue 285 with alanine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with COL1A1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with alanine at codon 285 of the COL1A1 protein (p.Pro285Ala). The proline residue is highly conserved and there is a small physicochemical difference between proline and alanine.
