Uncertain significance for Hermansky-Pudlak syndrome 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012388.4(BLOC1S6):c.79C>T (p.Pro27Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 27 of the BLOC1S6 protein (p.Pro27Ser). This variant is present in population databases (rs138284359, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with BLOC1S6-related conditions. ClinVar contains an entry for this variant (Variation ID: 565888). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
