Uncertain significance for Combined immunodeficiency due to ZAP70 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001079.4(ZAP70):c.827C>T (p.Thr276Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with methionine at codon 276 of the ZAP70 protein (p.Thr276Met). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and methionine. This variant is present in population databases (rs199502727, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with ZAP70-related conditions. ClinVar contains an entry for this variant (Variation ID: 565808). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The methionine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
