Uncertain significance for Familial acute necrotizing encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006267.5(RANBP2):c.1918-1G>A, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in RANBP2 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with RANBP2-related disease. This sequence change affects an acceptor splice site in intron 13 of the RANBP2 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency).
