NM_177438.3(DICER1):c.406G>C (p.Glu136Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E136Q variant (also known as c.406G>C), located in coding exon 3 of the DICER1 gene, results from a G to C substitution at nucleotide position 406. The glutamic acid at codon 136 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
