This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 1385, where C is replaced by T; at the protein level this means replaces serine at residue 462 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces serine with phenylalanine at codon 462 of the NBN protein (p.Ser462Phe). The serine residue is weakly conserved and there is a large physicochemical difference between serine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions.
