This variant lies in the NF1 gene (transcript NM_001042492.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3315, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 25 of the NF1 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NF1-related disease. Experimental studies have shown that a different change at the same nucleotide (c.3315-1G>T) results in skipping of exon 26 and a frameshift (p.Tyr1106Leufs*28) that is predicted to lead to an absent or disrupted protein product (PMID: 23913538).
