This variant lies in the KAT6B gene (transcript NM_012330.4) at coding-DNA position 3118, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 1040 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q1040X variant in the KAT6B gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Furthermore, the Q1040X variant is not observed in large population cohorts (Lek et al., 2016). We interpret Q1040X as a pathogenic variant.
