This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 2767, where C is replaced by A; at the protein level this means replaces leucine at residue 923 with isoleucine — a missense variant. Submitter rationale: This variant is denoted NF1 c.2767C>A at the cDNA level, p.Leu923Ile (L923I) at the protein level, and results in the change of a Leucine to an Isoleucine (CTA>ATA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. NF1 Leu923Ile was not observed in large population cohorts (Lek 2016). This variant is located in the GTPase activating protein domain (Xu 1990, Luo 2014). In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function. Based on currently available evidence, it is unclear whether NF1 Leu923Ile is a pathogenic or benign variant. We consider it to be a variant of uncertain significance.
