NM_001042492.3(NF1):c.5442C>G (p.His1814Gln) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1793Q variant (also known as c.5379C>G), located in coding exon 37 of the NF1 gene, results from a C to G substitution at nucleotide position 5379. The histidine at codon 1793 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
