This variant lies in the SOS1 gene (transcript NM_005633.4) at coding-DNA position 25, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 9 with lysine — a missense variant. Submitter rationale: The E9K variant in the SOS1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E9K variant is not observed in large population cohorts (Lek et al., 2016). The E9K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, in silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. We interpret E9K as a variant of uncertain significance.
