Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.382G>A (p.Val128Met), citing Ambry Variant Classification Scheme 2023: The p.V128M variant (also known as c.382G>A), located in coding exon 4 of the DNAH5 gene, results from a G to A substitution at nucleotide position 382. The valine at codon 128 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
