Pathogenic for Glycine encephalopathy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000170.3(GLDC):c.2919+1G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: GLDC c.2919+1G>A is located in a canonical splice-site within the last intron and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of GLDC function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. The variant allele was found at a frequency of 1.6e-05 in 250480 control chromosomes. c.2919+1G>A has been reported in the literature as a biallelic genotype in multiple individuals affected with Glycine Encephalopathy (Non-Ketotic Hyperglycinemia)(e.g. Coughlin_2017). These data indicate that the variant is very likely to be associated with disease. The following publication has been ascertained in the context of this evaluation (PMID: 27362913).
