Likely pathogenic for Glycine encephalopathy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000170.3(GLDC):c.2838+5G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: GLDC c.2838+5G>A alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Four predict the variant weakens a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 250928 control chromosomes. c.2838+5G>A has been reported in the literature in the compound heterozygous state in at least four individuals affected with Glycine Encephalopathy (Non-Ketotic Hyperglycinemia) (e.g. Conter_2006, Swanson_2015, Coughlin_2017, internal data). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
