This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 2506, deleting one base; at the protein level this means shifts the reading frame starting at serine residue 836, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ser836Valfs*11) in the FBN1 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has been observed in an individual with clinical features of Marfan syndrome (PMID: 30048161). ClinVar contains an entry for this variant (Variation ID: 560217). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
