Submitter rationale: The c.22696-1G>A variant in NEB has not been previously reported in the literature in individuals with nemaline myopathy, but has been identified in 0.0008% (9/1158188) of European (non-Finnish) chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP ID: rs745511810). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (Variation ID: 558699) and has been interpreted as likely pathogenic by Counsyl and Labcorp Genetics. This variant is located in the 3' splice region. Computational tools predict a splicing impact, though this information is not predictive enough to determine pathogenicity. There is an in-frame cryptic splice site 6 bases from the intron-exon boundary, providing evidence that this variant may delete 2 amino acids instead of causing loss of function. However, this information is not predictive enough to determine pathogenicity. Loss of function of the NEB gene is an established disease mechanism in autosomal recessive nemaline myopathy.
