Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.5570C>G (p.Ser1857Cys), citing Ambry Variant Classification Scheme 2023: The p.S1858C variant (also known as c.5573C>G), located in coding exon 8 of the ALMS1 gene, results from a C to G substitution at nucleotide position 5573. The serine at codon 1858 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
