Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_007294.4(BRCA1):c.900A>C (p.Glu300Asp), citing Quest Diagnostics criteria: The BRCA1 c.900A>C (p.Glu300Asp) variant has been reported in the published literature in an individual with ovarian cancer (PMID: 33888336 (2021)), and described to be located in a region of the BRCA1 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). It has also been predicted to affect gene function and deleterious from comparative evolutionary and ancestral methods, and described to be a conservative substitution at a fixed site (PMIDs: 12531920 (2003), 15385441 (2004) and 16518693 (2006)). The frequency of this variant in the general population (Genome Aggregation Database, http://gnomad.broadinstitute.org) is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
