Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251416 control chromosomes (gnomAD v2.1). c.2578G>C has been observed in two unrelated individuals affected with Glycine Encephalopathy (Non-Ketotic Hyperglycinemia), who both carried a (likely) pathogenic variant in trans (Coughlin_2017). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
