This variant lies in the PTS gene (transcript NM_000317.3) at coding-DNA position 400, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 134 with lysine — a missense variant. Submitter rationale: Variant summary: PTS c.400G>A (p.Glu134Lys) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 250752 control chromosomes (gnomAD). c.400G>A has been reported in the literature in individuals affected with 6-Pyruvoyl-Tetrahydropterin Synthase Deficiency (examples: Khatami_2017, Almannai_2019, Gundorova_2021). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
