This variant lies in the EVC gene (transcript NM_153717.3) at coding-DNA position 364, where C is replaced by T; at the protein level this means replaces proline at residue 122 with serine — a missense variant. Submitter rationale: Variant summary: EVC c.364C>T (p.Pro122Ser) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251482 control chromosomes. c.364C>T has been reported in the literature in the compound heterozygous state in at least one individual affected with Ellis-van Creveld syndrome (D'Asdia_2013). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
