Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000203.5(IDUA):c.1154C>G (p.Pro385Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: IDUA c.1154C>G (p.Pro385Arg) results in a non-conservative amino acid change located in the Glycosyl hydrolases family 39, N-terminal catalytic domain (IPR049166) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 158834 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1154C>G has been reported in the literature in the compound heterozygous state in at least one individual affected with Mucopolysaccharidosis Type 1 and in one individual with an abnormal newborn screen who had normal glycosaminoglycans (Bertola_2011, Fillman_2023). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
