This variant lies in the ACADS gene (transcript NM_000017.4) at coding-DNA position 431, where C is replaced by T; at the protein level this means replaces threonine at residue 144 with isoleucine — a missense variant. Submitter rationale: Variant summary: ACADS c.431C>T (p.Thr144Ile) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251340 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.431C>T has been reported in the literature (example: Huang_2016). This report however, does not provide unequivocal conclusions about association of the variant with Deficiency Of Butyryl-CoA Dehydrogenase. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
