NM_013339.4(ALG6):c.1390C>T (p.Gln464Ter) was classified as Pathogenic for ALG6-congenital disorder of glycosylation 1C by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the ALG6 protein in which other variant(s) (p.Ser478Pro) have been determined to be pathogenic (PMID: 10914684). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 555315). This variant has not been reported in the literature in individuals affected with ALG6-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln464*) in the ALG6 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 44 amino acid(s) of the ALG6 protein.
