This variant lies in the USH2A gene (transcript NM_206933.4) at coding-DNA position 43, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 15 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Variant summary: USH2A c.43C>T (p.Gln15X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 250392 control chromosomes (gnomAD).
