NM_000317.3(PTS):c.216T>A (p.Asn72Lys) was classified as Pathogenic for 6-Pyruvoyl-tetrahydrobiopterin synthase deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): ClinVar contains an entry for this variant (Variation ID: 554191). This missense change has been observed in individual(s) with biopterin-deficient hyperphenylalaninemia and/or PTS-related conditions (PMID: 31332730, 33822819). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 72 of the PTS protein (p.Asn72Lys). For these reasons, this variant has been classified as Pathogenic. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
