Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.10630A>G (p.Arg3544Gly), citing Ambry Variant Classification Scheme 2023: The p.R3545G variant (also known as c.10633A>G), located in coding exon 16 of the ALMS1 gene, results from an A to G substitution at nucleotide position 10633. The arginine at codon 3545 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
