NM_007294.4(BRCA1):c.4240dup (p.Leu1414fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.4240dupC pathogenic variant in the BRCA1 gene has previously been reported in at least two individuals with breast cancer (Fackenthal et al., 2012). The c.4240dupC variant causes a frameshift starting with codon Leucine 1414, changes this amino acid to a Proline residue and creates a premature Stop codon at position 14 of the new reading frame, denoted p.Leu1414ProfsX14. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.4240dupC variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.4240dupC as a pathogenic variant.
