Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.4057G>T (p.Glu1353Ter), citing Ambry Variant Classification Scheme 2023: The p.E1353* pathogenic mutation (also known as c.4057G>T), located in coding exon 9 of the BRCA1 gene, results from a G to T substitution at nucleotide position 4057. This changes the amino acid from a glutamic acid to a stop codon within coding exon 9. This mutation was identified in one patient with high grade serous ovarian cancer (Weren et al. Hum. Mutat. 2017 02;38(2):226-235). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
