NM_000492.4(CFTR):c.3139+3A>G was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: CFTR c.3139+3A>G alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a canonical 5' donor site and one predicts the variant has no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250706 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3139+3A>G has been observed in individuals affected with pancreatitis (example: Audrezet_2008, Masson_2013). These reports do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
