Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.10625A>G (p.Tyr3542Cys), citing Ambry Variant Classification Scheme 2023: The p.Y3543C variant (also known as c.10628A>G), located in coding exon 16 of the ALMS1 gene, results from an A to G substitution at nucleotide position 10628. The tyrosine at codon 3543 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
