This variant lies in the ERCC6 gene (transcript NM_000124.4) at coding-DNA position 2038, where A is replaced by G; at the protein level this means replaces asparagine at residue 680 with aspartic acid — a missense variant. Submitter rationale: Variant summary: ERCC6 c.2038A>G (p.Asn680Asp) results in a conservative amino acid change located in the Helicase superfamily 1/2, ATP-binding domain (IPR014001) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251396 control chromosomes. c.2038A>G has been reported in the literature in individuals affected with Cockayne Syndrome (Laugel_2010, Calmels_2018). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The DNA translocase activity of the orthologous variant in yeast was approximately 10%-<30% of the normal protein, measured by an in vitro yeast chromatin remodeling assay (Yan_2021).
