This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 3583, deleting one base; at the protein level this means shifts the reading frame starting at histidine residue 1195, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Variant summary: BRCA1 c.3583delC (p.His1195IlefsX15) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 250844 control chromosomes (gnomAD). c.3583delC has been reported in the literature in individuals affected with Hereditary Breast and Ovarian Cancer (e.g. Esteban Cardenosa_2008, Rebbeck_2018). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters including an expert panel (ENIGMA) (evaluation after 2014) cite the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
