NM_006941.4(SOX10):c.334A>G (p.Met112Val) was classified as Pathogenic for Sensorineural hearing loss disorder; Abnormal inner ear morphology; Blue sclerae; Micropenis; Waardenburg syndrome type 4C by Department of Otolaryngology-Head and Neck Surgery, Shanghai Jiao Tong University Affiliated Sixth People’s Hospital, citing ACMG Guidelines, 2015: NM_006941.4(SOX10):c.334A>G (p.Met112Val) is a parentally confirmed de novo variant (PS2) located within the conserved HMG functional domain of SOX10 (PM1), and pathogenic substitutions at the identical amino acid residue (NM_006941.4(SOX10):c.336G>A (p.Met112Ile), VCV001065836.5) have been previously documented (PM5). The variant is rare in general populations (PM2_Supporting) and predicted damaging by multiple bioinformatic algorithms (PP3). The patient’s phenotype of profound congenital deafness, inner ear developmental malformations, blue sclera and micropenis is consistent with Waardenburg syndrome type 4 (PP4). This variant is classified as Pathogenic following the 2015 ACMG/AMP guidelines and accounts for the proband’s clinical presentations.
