This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 4635, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 1545 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Y1524X nonsense variant has been reported previously in association with neurofibromatosis type 1 (Sabbagh et al., 2013). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016). We interpret this variant as pathogenic.
