Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_007294.4(BRCA1):c.3041T>A (p.Met1014Lys), citing ACMG Guidelines, 2015: This missense variant replaces methionine with lysine at codon 1014 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been detected in a breast cancer case-control meta-analysis in 6/60466 cases and 0/53461 unaffected individuals (PMID: 33471991; Leiden Open Variation Database DB-ID BRCA1_006366). A multifactorial analysis has reported a likelihood ratio for pathogenicity based on segregation of 1.9234 (PMID: 31131967). While there is a suspicion that this variant may be disease-causing, a study has reported that disease-causing missense variant in this exon is unlikely (PMID: 31911673). This variant has been identified in 6/251136 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
