Uncertain significance — the classification assigned by GeneDx to NM_030773.4(TUBB1):c.320C>T (p.Thr107Met), citing GeneDx Variant Classification (06012015). This variant lies in the TUBB1 gene (transcript NM_030773.4) at coding-DNA position 320, where C is replaced by T; at the protein level this means replaces threonine at residue 107 with methionine — a missense variant. Submitter rationale: The T107M variant in the TUBB1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is observed in 3/30782 (0.0097%) alleles from individuals of South Asian background and 10/277212 (0.0036%) global alleles in large population cohorts (Lek et al., 2016). The T107M variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret T107M as a variant of uncertain significance.
