Submitter rationale: The T151S variant in the UBE3A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The T151S variant is observed in 1/8724 (0.01%) alleles from individuals of African background in large population cohorts (Lek et al., 2016). The T151S variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. In silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. However, the majority of missense variants in the UBE3A gene are considered pathogenic (Stenson et al., 2014). We interpret T151S as a variant of uncertain significance.
