This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 2643, deleting one base; at the protein level this means shifts the reading frame starting at glutamic acid residue 881, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2643delA pathogenic mutation, located in coding exon 9 of the BRCA1 gene, results from a deletion of one nucleotide at nucleotide position 2643, causing a translational frameshift with a predicted alternate stop codon (p.E881Dfs*12. This alteration was identified in an individual diagnosed with ovarian cancer (Gorodnova TV et al. Int J Gynecol Cancer, 2018 10;28:1498-1506). Additionally, this alteration was identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
